abstract | The present invention relates to an antibacterial susceptibility testing composition comprising a microbial nuclease-detecting agent and a use thereof. The microbial nuclease-detecting agent is a FRET-DNA probe. When antibacterial susceptibility testing for a specific microbe is conducted with the agent according to the FRET-DNA probe-based AST (F-AST) method, bacterial cells are lysed and total proteins including nucleases are released from the lysate as a result of antibiotic-induced bacterial lysis. In this condition, the F-AST probe is cleaved by the nucleases to separate the quencher distant from the fluorophore, with the consequent significant increase of fluorescence signals, whereby the antibacterial susceptibility of microbes can be investigated accurately, rapidly, and easily. Therefore, the F-AST system of the present invention can be easily conducted and is fast and economically advantageous. Furthermore, being designed to be suitable for reading on a microtitration plate reader, the system can readily utilize conventional techniques and provides a basis for clinical therapy. |
