abstract Disclosed are a culture system and method for expanding hematopoietic stem cells and the use thereof. The culture system comprises a cell culture medium containing sodium butyrate. The method comprises obtaining hematopoietic stem cells in a fresh blood sample to use as sample cells and using the cell culture medium containing sodium butyrate for in vitro expansion culture to obtain hematopoietic stem cells. The beneficial effects: 1. Total number of the cells obtained by in vitro expansion of umbilical cord blood hematopoietic stem cells is increased by adding sodium butyrate, providing a preliminary technical foundation for clinical trials; 2. The hematopoietic stem cells following expansion comprise a higher proportion of CD34 + CD90 + cell populations, and possess a even better CFU colony forming ability, suggesting that the hematopoietic stem cells are more primitive and have a stronger differentiation potential to rebuild the blood system and can effectively support clinical treatment needs; 3. In vitro expansion of the hematopoietic stem cells becomes easier, safer and more efficient.
