The invention provides a novel artificially-designed antibody molecule, comprising (i) a single-domain antigen binding site; (ii) an antigen-binding Fab fragment, wherein (i) is located at an N-terminal of a light chain variable domain (VL) of (ii) or at a C-terminal of a light chain constant region (CL) of (ii), or (i) is located at an N-terminal of a heavy chain variable domain (VH) of (ii) or at a C-terminal of an immunoglobulin CH1 domain of (ii), (i) and (ii) respectively bind to the same or different antigens, and a linker peptide may or may not exist between (i) and (ii); and (iii) an immunoglobulin Fc domain located at the C-terminals of (i) and (ii). The invention also provides a polynucleotide encoding the antibody molecule, a vector containing the polynucleotide, a host cell containing the polynucleotide or the vector, an immunoconjugate containing the antibody molecule and a pharmaceutical composition containing the antibody molecule, and a use of the antibody molecule in immunotherapy, or prevention and/or diagnosis of a disease. |
