Provided are a chimeric antigen receptor, a NKG2D CAR-NK cell expressing the chimeric antigen receptor, a preparation method therefor and the use thereof. The chimeric antigen receptor comprises an antigen binding domain, a transmembrane domain and a co-stimulatory signaling region. The antigen binding domain can specifically bind to a tumor-specific antigen NKG2D ligand, and activate NK cells through the transmembrane domain and the co-stimulatory signaling region. The CAR-NK cell provided by the present invention uses the NKG2D ligand as a target antigen, and specifically kills tumor cells using NKG2D CAR-NK cells. The CAR-NK cell can be used as a medicament for treating tumor diseases and can be used for treating the tumor in which the ligand of NKG2D molecules highly expresses, providing a new method for the prevention and treatment of tumors. |
