Disclosed are an improved promoter and the use thereof, wherein the improvement is mutating the nucleic acid sequence between the -35 region and the -10 region in the promoter region into an endonuclease recognition site. This improvement is used for overcoming the problem that transcription or translation products of an exogenous gene under a strong promoter may be toxic to the host and may thereby result in the inability to clone, and for avoiding the phenomena of false positives and false negatives during blue-white screening. |
