abstract | The present invention provides methods and compositions for analysis using digital amplification tests with unconventional / inverse photoluminescence changes to indicate the presence of one or more targets. In an exemplary method, isolated volumes can be formed, only a subset of these containing a target. Each volume may include a probe having a label, a heatsink with a fire extinguisher and designed to hybridize with the probe to extinguish the label, and a splitter designed to hybridize with the probe and / or the heatsink to block the hybridization of the dissipator with the probe. An amplification reaction can be performed in the volumes to generate an amplicon corresponding to the target. The separator can hybridize with the amplicon and can be expanded or degraded by the amplification reaction. The photoluminescence of the label can be detected from the volumes, and the photoluminescence of target containing volumes can be lower than that of the non-target volumes. |
