abstract | The invention relates to non-naturally occurring tRNAsec and methods of use thereof for the recombinant expression of genetically engineered proteins to include one or more selenocysteine residues. The non-naturally occurring tcRNAs can be used for the recombinant manufacture of selenocysteine-containing polypeptides encoded by mRNA without the need for a SECIS element. In some embodiments, the selenocysteine-containing polypeptides are made by co-expressing non-naturally occurring tcRNAs in a recombinant expression system, such as E. coli, with SerRS, EF-Tu, SelA or PSTK and SepSecS, and an mRNA having at least one codon that recognizes the tRNAs anticodon of unnatural origin. |
