abstract The present invention provides isolated and modified ADAM10 modulating peptides and methods of use thereof for modulating the biological activities of ADAM10 or inhibiting the biological activities of ADAM10 associated with diseases, disorders or conditions in subjects, with the result that for example, decreasing inflammation and inhibiting unwanted cell proliferation. In some embodiments, ADAM10 modulated isolated and modified peptides are based on SEQ ID NO: 3 or SEQ ID NO: 4 and, in some embodiments, include modifications at or near the N- and / or N-terminus. or C-termini of the described peptides, as well as substitutions, insertions, and deletions at one or more amino acid positions of the ADAM10 prodomain peptides described herein.
