abstract | The present invention provides markers, conjugates, compositions and methods for imaging, mapping and hypoxia therapy. A compound comprising a bio-reductive activation arm (BA), a linker arm and a mapping click, BA containing one or more of the substituted or unsubstituted nitroimidazoles in 2/4/5 or benzotriazene-1,4 substituted dioxides or substituted 1,2,3 / 1,2,4-triazoles or substituted 1,4-benzoquinones or a combination of two homo-BA or hetero-BA fragments, the linker arm containing alkane, alkene linkers alkyne, alicyclic or aromatic C1-16 compounds, with or without heteroatoms as in ethers, amines, esters, acids, amides, 5- and 6-membered sugar rings with the substitution as described above , both monosaccharides and disaccharides, the mapping click containing one of -N3-, CN, SCN, substituted or unsubstituted, substituted alkynes for click chemistry. Said compound undergoes a click-in-situ reaction after its distribution into cells or tissues to bind to a reporter group that may be non-chelated or chelated by a radioactive or non-radioactive metal or a radiohalogen for PET / SPECT or a dye for a fluorescent / optical reporting group, thus performing hypoxia imaging. |
