abstract | The present invention relates to methods for use in virus inactivation. Virus inactivation methods with N-methylglucamides are applicable to the purification process of biologically active drugs such as protein subunits, proteins (enzymes, factors, etc.), recombinant proteins, antibodies, vaccines or gene therapy products. The detergents used in this process are based on multiple homologs of N-methylglucamide, consisting of a hydrophilic glucose moiety and a hydrophobic fatty acid tail, linked by an amide bond. In addition, these sugar-based detergents are non-ionic in nature, ie, they do not disrupt drug protein, plasma biology, non-enveloped viral vaccine, or viral particles associated with adenoviruses. The invention also relates to a method of purifying a biological product solution of interest having an unidentified enveloped virus contaminant, comprising incubating a biological product solution of interest with a standard solution, inactivation of any potential enveloped virus contaminant present in the biological product solution of step (a), measuring the inactivated virus present in the final solution of step (b), incubating a solution of biological product of interest with a solution of N-methylglucamide, the measurement of the inactivated virus present in the final solution of step (d), and the comparison of the results of the final solutions of step (c) and the step ( e). |
