The invention relates to a composition comprising (a) an inducible transgene construct comprising a sequence encoding an inducible promoter and a sequence encoding a transgene, and (b) a receptor construct having a coding sequence for a transgene constitutive promoter and a coding sequence for an exogenous receptor, the invention being characterized in that upon integration of the construct (a) and the construct (b) into a genomic sequence of a cell, the Exogenous reporter is expressed, and in that the exogenous reporter, upon binding to a ligand, transduces an intracellular signal that targets the inducible promoter of (a) to modify gene expression. The invention also provides methods of introducing compositions into cells and using the resulting cells in adoptive cell therapies. |
