Nanocapsules comprising a unique ribonucleoprotein complex (RNP) as a core and a biodegradable crosslinked polymer shell that encapsulates the core, wherein the RNP complex comprises a Cas9 polypeptide and a guide RNA, and the biodegradable crosslinked polymeric shell comprises polymerized monomers of imidazolyl- (meth) acryloyl monomeric monomers, bisacryloyl disulfide monomers (biodegradable crosslinking agent), optionally PEG- (meth) acryloyl monomers, and either cationic (meth) acryloyl monomers, (meth) acryloyl monomers, anionic, either cationic and anionic (meth) acryloyl monomers (optionally in combination with nonionic (meth) acryloyl monomers) as defined in the description. Processes for preparing said nanocapsules, kits containing them, and methods of administering encapsulated RNPs to cells are further described. |
