abstract The present invention provides compositions and methods for modifying genomic DNA sequences. The methods produce double-strand breaks (DSB) at predetermined target sites in a genomic DNA sequence, resulting in mutation, insertion and / or deletion of DNA sequences at the site (s) ( s) target (s) in a genome. The compositions comprise DNA constructs comprising nucleotide sequences that encode a Csm1 protein operably linked to a promoter that can be used in the cells of interest. DNA constructs can be used to direct the modification of genomic DNA at predetermined genomic loci. The invention also relates to methods of using these DNA constructs to modify genomic DNA sequences. In addition, the invention provides compositions and methods for modulating gene expression. The compositions comprise DNA constructs comprising a promoter that can be used in cells of interest operably linked to nucleotide sequences that encode a mutated Csm1 protein that no longer has the potentially bounded DSB production capacity. to a domain that regulates transcriptional activity. The methods can be used to upregulate or downregulate gene expression at predetermined genomic loci.
