abstract The present invention relates to a method for synthesizing compounds useful in the preparation of antibody-drug conjugates (ADC), namely dimeric bifunctional dimeric prodrugs based on duocarmycin analogs. In another aspect, the invention relates to compounds obtained by the process according to the present invention. The monoprotected difunctional prodrug is used to prepare antibody-drug conjugates composed of an antibody fragment and the monoprotected difunctional prodrug. The invention also relates to the conjugates of antibody compounds thus obtained. The invention further relates to a method for preparing an antibody-drug conjugate composed of two identical or different antibody fragments, as well as the antibody compound conjugate containing two different antibody fragments. Accordingly, the conjugates can be used in pharmaceutical compositions, particularly for use in the treatment of tumors, for example for use in ADC therapy.
