The present invention provides a donor construct, a gene inactivation method, and an associated system and kit. The donor construct is a linear donor DNA or may be cleaved in a cell to produce the linear donor DNA. The gene inactivation method utilizes a marker gene contained in the donor construct to enrich cells in which a gene is inactivated to enhance the gene inactivation generation efficiency using a sequence-specific nuclease. |
