abstract The present invention provides methods of suppressing the formation of adapter dimers comprising: providing a target polynucleotide having a 5 'end and a 3' end; the use of a double-stranded DNA adapter with a 5 'end and a 3' end which have complementary sequences from each other, ligation of the double-stranded adapter to the target polynucleotide to form a ligation product . The invention also relates to a method for preparing a library of nucleic acid sequences comprising: providing a double-stranded DNA adapter with 5 'and 3' ends having sequences complementary to each other contacting the adapter with target nucleic acid sequences having a 5 'end and a 3' end, and ligating the adapter with a sequence complementary to the 5 'and 3' ends of the target nucleic acid using a double-stranded DNA ligase. The invention also relates to kits for suppressing adapter dimer formation in deep sequencing containing a double-stranded DNA adapter with 5 'and 3' ends having sequences complementary to one another, appropriate enzymes, buffers, dNTPS, etc.
