The present invention relates to plasmid DNA expressing CRISPR and a gene delivery system targeted to hepatocytes infected with the hepatitis B virus. The invention also relates to methods for orienting the formation of a CRISPR system in these cells to ensure improved specificity of target recognition to modify, improve or treat chronic HBV infection. In addition, the CpG content of the expression plasmid DNA was minimized to reduce inflammation and maximize gene expression. |
