abstract | The subject of the present invention relates to a double-stranded nucleic acid for silencing the expression of a gene coding for a PRODH / POX protein in a cell, an expression vector for silencing the expression of a gene coding for a PRODH / POX protein comprising said double-stranded nucleic acid, a host cell comprising said double-stranded nucleic acid or vector, a cell clone having a silent expression of a gene coding for the PRODH / POX protein comprising said double nucleic acid strand or vector, a pharmaceutical composition for silencing the expression of a gene encoding a PRODH / POX protein in an organism comprising said double-stranded nucleic acid or vector, and a pharmaceutically acceptable carrier or diluent. The subject of the invention further relates to an in vitro method of silencing the expression of a gene coding for a PRODH / POX protein in a cell, in which a double-stranded nucleic acid or a vector is introduced, and of maintaining the cell containing the double-stranded nucleic acid or introduced vector under conditions and for a period of time sufficient for silencing the expression of a gene encoding the PRODH / POX protein in a cell. The use of the double-stranded nucleic acid according to the invention in therapy, for treating and / or preventing a disorder or a disease characterized by a metabolism of the altered proline, preferably a neoplastic disease, in particular breast cancer, the Marfan syndrome, Ehlers-Danlos syndrome and osteogenesis imperfecta; to inhibit the growth and / or proliferation of a neoplastic cell; to induce apoptosis and / or autophagy; its use in diagnostics, in particular for diagnosing diseases characterized by an altered proline metabolism, preferably a neoplastic disease, in particular breast cancer, Marfan syndrome, Ehler-Danlos syndrome, and osteogenesis imperfecta; its use in the manufacture of a cell line with silent expression of a gene coding for the PRODH / POX protein; its use in the manufacture of a transgenic non-human organism with silent expression of a gene coding for the PRODH / POX protein; its use in the manufacture of a silent expression cell clone of a gene coding for the PRODH / POX protein, preferably a clone of MCF-7 cells with a silent expression of a gene coding for the PRODH / POX protein are in further described.
