abstract |
The present invention relates to a protein complex in which an estrogen receptor 2 (ERT2) is fused to Cas 9 (CRISPR associated protein 9) and a recombinant vector carrying a gene encoding it, where ERT2 is bound to the N-terminus and at the C-terminus of Cas9 whose nuclear localization sequence (NLS) was deleted. The protein complex according to the present invention has the advantage of moving from the cytosol to the nucleus by translocation at a certain time point during treatment with tamoxifen. Thus, the protein complex that has translocated into the nucleus can modify a specific DNA using a guide RNA (gRNA), to finally allow a more elaborate DNA modification operation in a desired part at a point desired time. Therefore, the present invention should be used to develop a new practical DNA modification technology. |
