abstract A method for knocking out target genes in T cells in vitro based on the CRISPR-Cas9 system is provided. Also provided are crRNAs targeting the target genes TRAC, B2M and PD1, and sgRNA, Cas9 protein, oligodeoxyribonucleic acid (N-oligo) or fish sperm DNA fragments comprising the tracrRNA junction of the crRNA and the Cas9 protein. A kit for knocking out TCR, B2M and/or PD1 genes in T cells. Also provided are gene knockout T cells obtained according to the methods of the invention and uses thereof.
