titleOfInvention Mutant gaussia princeps luciferase having amplified bioluminescence intensity
abstract The present invention relates to a Gaussia princeps luciferase mutant which is modified by site-specific mutagenesis to amplify the bioluminescence intensity of Gaussia princeps -derived luciferase. More particularly, the present invention relates to a single- or multiple-mutation Gaussia princeps luciferase prepared by substituting at least one of amino acids at positions 60, 88, 89, 90, and 103 of the amino acid sequence of Gaussia princeps Luciferase ( G Luc) with a different amino acid, a polynucleotide encoding the mutant sequence, a recombinant vector carrying the polynucleotide, a transformant anchoring the recombinant vector, a method for preparing a mutant Gaussia princeps luciferase improved in bioluminescence intensity from the transformant, and a kit comprising the mutant Gaussia princeps luciferase. The present invention can prepare a mutant Gaussia princeps luciferase improved in bioluminescence signal by mutating an amino acid residue playing a critical role in amplifying the bioluminescence signal of Gaussia princeps luciferase and can usefully apply the same to cell monitoring and drug screening.
