abstract The present invention provides an RNA comprising in the 5' to 3' direction a first sequence and a second sequence, the first sequence being spaced apart from the second sequence by a spacer sequence, the first and second sequences being substantially complementary and forming a duplex via Watson and Crick base pairing, the second sequence being able to hybridise with the mRNA of a target gene in an organism, the RNA being substantially in the form of a hairpin, the second sequence being fully complementary to the said mRNA along the region at which it hybridises, the first sequence comprising nucleotide bases which do not Watson and Crick base pair with the corresponding bases in the second sequence of the duplex wherein the duplex does not comprise regions of 3 or more consecutive mismatched bases or an RNA comprising in the 5' to 3' direction a first sequence and a second sequence, the first sequence being spaced apart from the second sequence by a spacer sequence, the first and second sequences being substantially complementary and forming a duplex via Watson and Crick base pairing, the first sequence being able to hybridise with the mRNA of a target gene in an organism, the RNA being substantially in the form of a hairpin, the first sequence being fully complementary to the said mRNA along the region at which it hybridises, the second sequence comprising nucleotide bases which do not Watson and Crick base pair with the corresponding nucleotide bases in the first sequence of the duplex, wherein the duplex does not comprise regions of 3 or more consecutive mismatched bases.
