abstract | A method for simultaneously detecting a single cell mRNA expression level and telomere length, said method comprising: (1) single cell capture and lysis; (2) single cell mRNA and gDNA separation; (3) single cell mRNA amplification and telomere-specific sequence amplification; (4) qPCR detection of single cell gene expression; (5) detection of single cell relative telomere length. The method effectively lyses the single cell, releases the complete mRNA and gDNA, uses a biotin-labeled primer to capture the mRNA, uses magnetic beads to absorb the biotin, physically separates the mRNA and gDNA, and respectively detects the single cell gene expression and length. The method obtains a gene expression level and telomere length of a same cell, and performs correlation analysis. The detection method has high accuracy and sensitivity, is simple and convenient, and performs parallel gene expression and telomere length detection on a variety of mammalian cells at the single cell level. |
