titleOfInvention Method for analyzing sialyl sugar chain
abstract The purpose of the present invention is to carry out the analysis of the structure of a sialyl sugar chain, including the mode of the linkage of sialic acid bound to a terminal of a sugar chain, with high efficiency.nA sialic acid residue contained in a sample (a sugar chain) of interest is modified in a linkage-mode-specific manner (S2), and subsequently the sample is subjected to mass spectrometry to obtain mass spectra (S3). The monosaccharide composition is estimated in a round-robin manner using the types and the range of number of monosaccharides as conditions on the basis of the mass of a modified sugar chain which is estimated from the m/z value of each of peaks observed on the mass spectra (S4), and subsequently candidates for the monosaccharide composition which are respectively nominated for the peaks are narrowed down using the difference in mass of modified forms produced by linkage-mode-specific modification (S5). More specifically, peaks having difference in mass, which corresponds to the difference in mass of the modified forms produced by linkage-mode-specific modification, are found and are deemed as a group of peaks which is derived from a single sugar chain including a linkage isomer, and the narrowing down is carried out by eliminating candidates which do not meet the conditions, such as the presence or absence of a sialic acid residue modified form or the number of the sialic acid residue modified form, from the candidates for the monosaccharide composition which correspond to the peaks belonging to the above-mentioned group.
