titleOfInvention Additive to nutrient medium for forming bacterial biofilms. variants
abstract The invention relates to microbiology and biotechnology, and may be used for diagnostic purposes, and also in biotechnological production for accumulating a microbe biomass. Three variants of additives are proposed: 1. An additive to a nutrient medium for forming bacterial biofilms, including a fibrin biopolymer in a quantity of 0.05 grams to 10 grams per 1 kilogram of nutrient medium, and a proteinase inhibitor in a quantity of 0.002 grams to 0.5 grams per 1 kilogram of nutrient medium. 2. An additive to a nutrient medium for forming bacterial biofilms, including fibrinogen in a quantity of 0.05 grams to 10 grams per 1 kilogram of nutrient medium, plasma-coagulase having a ratio to fibrinogen of 1:128 to 1:10000 (mass:mass), and a proteinase inhibitor in a quantity of 0.002 grams to 0.5 grams per 1 kilogram of nutrient medium. 3. An additive to a nutrient medium for forming bacterial biofilms, including fibrinogen in a concentration of 0.01 grams/liter to 10 grams/liter, a "thrombin-ion Ca 2+ " complex containing thrombin having a ratio to fibrinogen of 1:10-1:500 and Ca 2+ ions in a quantity of 0.08 grams to 0.2 grams per 1 kilogram of nutrient medium, and a proteinase inhibitor in a quantity of 0.002 grams to 0.5 grams per 1 kilogram of nutrient medium. The proposed additives are universal, and can be introduced into the compositions of various nutrient media intended for cultivating various forms of bacteria with the aim of modeling biofilms. The use of the proposed additive allows for producing a biofilm which is based on strains of bacteria which are incapable of forming biofilms on known nutrient media without the proposed additive. The preparation of the additive is technologically feasible for mass production. Media containing same in the compositions thereof may be sterilized using conventional methods, for instance via filtration, have a sufficient storage life without losing useful properties, and do not require special conditions for disposal following use.
