abstract | The invention provides a method of biosynthesis of a cyclic peptide by enzymatically transforming a substrate peptide into the cyclic peptide, wherein the substrate peptide is expressed by a displaying genetic package and the enzyme is an enzyme-leader-fusion protein (ELF) which is a post-translationally modifying enzyme (PTME) that is fused to a cognate PTME-leader peptide, thereby obtaining the displaying genetic package carrying the cyclic peptide; and further provides a library of immobilized cyclic peptides, each with a length of at least 10 amino acids, obtainable by a method wherein a repertoire of variant substrate peptides comprising a randomised sequence is transformed by at least two ELFs which differ in the class of the PTMEs, which library comprises a variety of at least 10 6 library members, which variety comprises at least one of a) a different type of a heteroatom bridge linking amino acid side chains of the substrate peptide thereby forming a loop within the substrate peptide; b) a different loop position; or c) a different number of loops. |
