The present invention provides a buffer system and a method of using the buffer system to measure total alkaloid in tobacco or tobacco products through continuous flow. The buffer system comprises a buffer solution A and a buffer solution B. The buffer solution A is prepared by the following steps: weighing 60.5 to 83.0 g, preferably 71.6 g, of disodium hydrogen phosphate and 10.1 to 20.2 g, preferably 15.2 g, of trisodium phosphate, placing the weighed substances into a beaker, dissolving the mixture with water, transferring the solution into a volumetric flask of 1 L, and then filling water into the volumetric flask until the water reaches the scale. The buffer solution B is prepared by the following steps: weighing 5.5 to 8.0 g, preferably 7.0 g, of p-aminobenzene sulfonic acid, 60.5 to 83.0 g, preferably 71.6 g, of disodium hydrogen phosphate, 5.0 to 7.0 g, preferably 6.2 g, of sodium dihydrogen phosphate, and 10.0 to 12.1 g, preferably 11.8 g, of sodium citrate, placing the weighed substances into a beaker, dissolving the mixture with water, transferring the solution into a volumetric flask of 1 L, and then filling water into the volumetric flask until the water reaches the scale. |
