abstract |
Provided is a method for simply and highly accurately detecting short-chain RNA. Provided is a nucleic acid detection method that includes steps (a)-(c): (a) a step for producing a long reverse transcript from a target RNA by using a reverse transcription primer to perform a reverse transcription reaction that uses the target RNA as a template, the reverse transcription primer having at the 5' terminus thereof a sequence that is non-complementary to the target RNA, (b) a step for producing a double-stranded DNA amplified fragment that includes a single-stranded region at at least one terminus by using two primers to perform a nucleic acid amplification reaction that uses the reverse transcript as a template, and (c) a step for hybridizing the single-stranded region of the double-stranded DNA amplified fragment and an oligonucleotide probe that is fixed upon a solid phase. |
