Methods for isolating lineage-specific cells |
abstract |
A method of isolating high purity lineage-specific cells is disclosed. An initial cell is manipulated to contain at least one selection marker gene and at least one surface marker gene. Both the selection marker gene and the surface marker gene are operably linked to at least one lineage-specific promoter. The initial cell is transformed into a plurality of differentiating cells, which contains at least one lineage-specific cell, wherein the selection marker and/or surface marker are expressed. The lineage-specific cell is isolated with high purity by culturing the differentiating cells in the presence of a selection agent and/or by enriching the lineage-specific cell with the surface marker. |
