abstract | The invention concerns a method for inducing targeted meiotic recombination in a eukaryotic host cell, by co-expression in said cell of a first chimeric protein ME comprising an effector domain DE capable of inducing the formation of double-strand breaks in the DNA during meiotic recombination and an interaction domain DI1 formed by the first partner of a protein-protein pair having selective affinity, and a second chimeric protein comprising an interaction domain DI2 formed by the second partner of said protein-protein pair having selective affinity and a targeting domain DC capable of binding selectively with a target sequence present in the DNA of said host cell. The protein-protein interaction between domains DI1 and DI2 allow a non-covalent complex to form between module ME and module MC, and double-strand breaks to be induced by effector domain DE near the sequence targeted by targeting domain DC. |
