abstract The present invention relates to a method for separating polypeptides or proteins tagged with histidine glues from a mixture of proteins (for example, a cell extract), by means of a system of two aqueous phases in which one of the components is PEG, and in which the protein tagged with polyhistidine is preferably distributed in the PEG-rich phase under specific conditions. The purification process can include one or more washing steps and is completed with an inversion in the distribution of the tagged polypeptide or protein, induced by a change in the pH or by the addition of choline, which leads to the recovery of said peptide in the opposite phase. The described method is simple, economically efficient, scalable and does not require heavy-metal ions; said method is therefore suitable for use in the industrial sector.
