abstract A reconstituted human liver tumor model in a mouse and methods for making the same are disclosed. The mouse model includes a chimeric liver comprising mouse liver cells and integrated human liver cells. The liver tumors are genetically human because they arise from primary human hepatocytes that are transplanted and engrafted into host mice. Prior to transplantation, the primary human hepatocytes are genetically modified to (i) reduce or eliminate the function of a tumor suppressor gene and (ii) contain a recombinant human oncogene.
