abstract |
Provided is a practical method for enzymatically synthesizing c-di-GMP with good productivity.nA diguanylate cyclase having the following physicochemical properties (A) to (F) is used: (A) the activity: the diguanylate cyclase can catalyze the reaction 2GTP → c-di-GMP; (B) the molecular weight: 19800 ± 2000; (C) the optimum pH: 7.3 to 9.4; (D) the optimum temperature: 35 to 60˚C; (E) the thermal stability: the residual activity is 90% or more when heated under the conditions of 50˚C and pH 7.8 for 60 minutes; and (F) the diguanylate cyclase contains a GGDEF domain and does not have an amino acid sequence KXXD in an i-site thereof. |
