abstract |
Provided in the present invention is a method for the construction of a plasma DNA sequencing library and a kit thereof. The method of the present invention comprises the following steps: extracting the plasma DNA; connecting the plasma DNA with the sequencing linker and purifying the ligation product; PCR amplifying the purified ligation product; and purifying the amplified PCR product to obtain the plasma DNA sequencing library, wherein the step of the 5'-terminal phosphorylation of the plasma DNA is excluded. End filling and end A-hanging of the extracted plasma DNA can be carried out after the plasma DNA extracting and prior to the connection of the plasma DNA with the sequencing linker. Also provided in the present invention is a corresponding kit for implementing the method above. |
