titleOfInvention Mosaic adenovirus production method
abstract The invention relates to a method for the production of mosaic hybrid adenovirus having the capsid of a first type of adenovirus and fibres of said first type and a second type. In particular, the invention relates to the production of adenovirus Ad5/40, comprising: i) infecting permissive cells, which are adapted for growth in suspension and which express fibre of the first type, with an adenovirus having a capsid of the first type and fibre of the second type, in the presence of a polycation (preferably polybrene); ii) culturing said cells; and iii) recovering the mosaic hybrid viral particles. Finally, the particles can be amplified into cells adapted for growth in suspension, which do not express the fibre of the first type of adenovirus, with the particles of the hybrid adenovirus being recovered. The invention also relates to kits and to the uses of said method.
