abstract The present invention relates to a method for preparing an aptamer coupled to a biological sample, wherein an aptamer library coupled to an analysis sample and an aptamer library coupled to a comparison sample to be compared with the analysis sample are prepared and the libraries are normalized and subtracted to prepare an aptamer more effective for the analysis of an analysis sample over a comparison sample. According to the present invention, the method for preparing an aptamer comprises the following steps: (A) preparing a starting oligonucleotide library; (B) preparing an aptamer library coupled to a biological analysis sample to be analyzed and an aptamer library coupled to a biological comparison sample to be compared with the analysis sample, respectively, from the starting oligonucleotide library; (C) normalizing and subtracting the aptamer libraries; and (D) obtaining the normalized and subtracted aptamer.
