abstract | The invention relates to a method for the light-microscopic imaging of a sample structure (2, 34), having the following steps: preparing the sample structure (2, 34) with markers which can be converted into a state that can be light-microscopically imaged; creating a sequence of individual image data sets by sequentially imaging the sample structure (2, 34) such that only a subset of all the markers is converted into the state that can be light-microscopically imaged for each image, wherein the markers of the respective subset have an averaged distance from each other, said distance being greater than the light-microscopic image resolution limit which determines the extension of a light distribution representing the correspondingly imaged marker; generating at least two data blocks, in each of which multiple successive individual image data sets are combined; superimposing the individual image data sets that are contained in the respective data block into a superimposed image data set; ascertaining an image offset between the superimposed image data sets; correcting the individual image data sets that are contained in at least one of the superimposed image data sets using the ascertained image offset; determining centers of gravity of the light distributions that represent the imaged markers; and combining the centers of gravity into an offset-corrected whole image. |
