abstract | The present invention provides a method of identifying a target peptide for quantification by mass spectrometry (MS), comprising: (i) mixing peptides from a pair of samples at y different ratios to produce y mixtures, wherein y is 2 or greater than 2; (ii) carrying out MS on each of the y mixtures to determine the ionic intensity of a peptide in each of the y mixtures; (iii) calculating the relative ionic intensity of said peptide in each of the y mixtures by comparing the ionic intensity of said peptide in each of the y mixtures to the maximum ionic intensity of said peptide across all of the y mixtures; and (iv) correlating the relative ionic intensity of said peptide in each of the y mixtures with the change in composition of the y mixtures; wherein if the relative ionic intensity of said peptide is directly proportional to the change in composition of the y mixtures, said peptide is a target peptide for quantification by MS. |
