The invention relates to an in vitro method for preparing an artificial tissue, to the resulting artificial tissue, to the pharmaceutical composition containing same and to the use of said tissue and composition for partially or fully increasing, restoring or replacing the functional activity of a tissue or a damaged organ, preferably the skin, bladder, urethra, cornea, oral mucosa, conjunctiva, abdominal wall, tympanic membrane, pharynx, larynx, intestine, peritoneum, ligament, tendon, bone, meninx or vagina. The method of the invention includes steps consisting in: adding fibrinogen to isolated cells, preferably to fibroblasts or keratocytes; adding an antifibrinolytic agent to the mixture; adding a coagulation factor, a source of calcium and thrombin; adding a polysaccharide, preferably agarose; culturing cells, preferably epithelial cells or stem cells from the umbilical cord, in or on the resulting product; and inducing the nanostructure of said product. The method also includes an intermediate step consisting in adding fibronectin after the fibrinolytic agent and another step in which collagen is added after the agarose. |
