abstract |
The present invention is generally related to a method of baculovirus expression of a LFn-fusion protein for use in delivering antigens across the membrane of a cell to elicit a strong response of cell-mediated immunity (CMI) to the antigen portion of the LFn-fusion protein. One aspect of the present invention relates to the methods of use of a baculovirus expression system to produce N-glycosylated forms of LFn and LFn fusion polypeptides, which are more effective than non-glycosylated forms of the fusion proteins for delivering antigens across the membrane of a eukaryotic cell to elicit a strong CMI. Another aspect relates to methods and compositions to elicit a CMI response in a cell to a target antigen, comprising contacting the cell with a target antigen fused to a LFn polypeptide, wherein the target antigen and/or the LFn polypeptide are N-glycosylated. |
