titleOfInvention Method for the production and purification of the factor derived from the pigmented epithelium of the retina in a yeast system
abstract The invention relates to a method for the production and purification of PEDF molecules and mutated and/or truncated versions of said molecule in a yeast expression system. The invention also relates to the protection of the PEDF molecules directly obtained and to the use of same in the preparation of pharmaceutical compositions. The method includes the following steps in which: the PEDF cDNA is obtained at least from cells of the retinal pigmented epithelium, mutant and/or truncated versions are obtained, the structures are cloned into a yeast expression vector, the yeast clones are transformed and selected, the yeast culture medium is inoculated with the clones obtained, the intact or truncated PEDF factor is induced in the yeast, the cells are eliminated from the culture by means of centrifugation, the culture medium containing the intact and/or truncated PEDF factor is changed, and said PEDF factor is purified and isolated.
