abstract The present invention relates to methods for introducing multiple transgenes into a primary cell as well as to methods for producing multi-transgenic primary cells, tissues and/or animals by utilizing nucleic acid constructs that are capable of de novo formation of artificial chromosomes. The present invention furthermore relates to uses of these multi-transgenic primary cells, tissues and animals, preferably in the context of xenotransplantation. The nucleic acid constructs capable of de novo forming artificial chromosomes are utilized as versatile and effective means for the transfer of multiple human transgenes and, thus, ultimately for the generation of animals as organ donors in xenotransplanation.
