abstract | The present invention describes the isolation, identification and characterization of a gene (gpgS) and of the function of the corresponding enzyme responsible for the synthesis of glucosyl-3-phosphoglycerate (GpgS), the precursor of glucosylglycerate, a component of the methylglucose polysaccharides from mycobacteria and some closely related bacteria. This gene is conserved among mycobacteria and is essential for the growth of Mycobacterium tuberculosis, the causative agent of tuberculosis. The invention discloses the gpgS gene, processes for production of the enzymes encoded by the genes from Mycobacterium bovis and Mycobacterium smegmatis, the properties of the enzymes, the construction of plasmids that include the genes and of Escherichia coli clones containing these plasmids and that can be used for the large scale production of the enzymes, aiming at the identification or development of specific molecules that can inhibit this enzyme involved in the metabolic pathway leading to the synthesis of the methylglucose polysaccharides from mycobacteria. Therefore the present invention is applicable to the area of the pharmaceutical industry. |
