abstract |
The present invention relates to Infectious Pancreatic Necrosis Virus (IPNV), the etiological agent of infectious pancreatic necrosis in salmonids that causes significant losses in the aquaculture sector. The viral capsid protein (VP2) gene was cloned into a yeast expression vector and expressed in Saccharomyces cerevisiae. Expression of the capsid gene in yeast resulted in the formation of about 20 sub-viral nanoscale particles composed solely of VP2 proteins. Anti-PNV antibodies were detected in rainbow trout vaccinated by injection of purified VP2 subviral particles (rVP2-SVP) or by ingestion of recombinant yeast expressing rVP2-SVP. The provocation test of a trout immunized with rVP2-SVP using a heterologous IPNV strain and the subsequent determination of the viral load showed that the fish vaccinated orally or by injection had lower DPNV loads than the native fish or whose vaccination has been simulated. This study demonstrates the ability of rVP2-SVP to induce a specific immune response and the ability of immunized fish to reduce viral load after experimentally induced IPNV infection. The invention is not limited to IPNV and is applicable to other similar viruses for which subviral particles can be made and administered to fish. |
