abstract The present invention relates to a knock-out vector produced using a reporter knock-in vector, a process for preparing the same, and a method for knocking out a gene from an animal cell. In particular, the present invention relates to a method for knocking out a function of a certain gene by replacing a knock-out gene, from the initiation codon, with a reporter gene ORF (Open Reading Frame) containing its initiation codon to construct a knock-out vector, and then introducing the knock-out vector into an animal cell, wherein LacZ, fluorescent proteins and the like are used as the reporter of the knock-out vector. The present invention, in knock-out vectors using LacZ, fluorescent proteins and the like as a reporter, replaces a knock-out gene with a reporter gene ORF containing its initiation codon from the initiation codon ATG of the knock-out gene. This invention thereby expresses a reporter more stably and effectively than the conventional IRES system.
