abstract |
It becomes possible to produce a mutant phosphoseryl-tRNA synthetase (SepRS) suitable for the method for the site-specific introduction of a phosphoserine into a protein, by analyzing the structure and the functions of SepRS derived from an archaebacterium. The mutant SepRS has an amino acid sequence having the substitution of at least one of glutamic acid residues at position-418 and position-420 and a threonine residue at position-423 by another amino acid residue in the amino acid sequence depicted in SEQ ID NO:2, and also has an improved binding affinity for suppressor tRNA compared to a wild-type SepRS having the amino acid sequence depicted in SEQ ID NO:2. |
