abstract |
Disclosed is a method for production of a protein having a non-natural type amino acid integrated therein. The method is characterized by employing either of a prokaryote-derived aaRS* or a eukaryote-type aaRS* and using any one of a prokaryotic cell and a eukaryotic cell as a host cell. The method comprises the steps of: (a) introducing both of a gene encoding a prokaryote-derived mutant aminoacyl tRNA synthetase having an increased specificity to a non-natural type amino acid that is similar to a given natural type amino acid compared to the natural type amino acid and a tRNA gene for a non-natural amino acid that can bind to the above-mentioned non-natural amino acid in the presence of the prokaryote-derived mutant aminoacyl tRNA synthetase into a prokaryotic cell capable of expressing both of a gene encoding a eukaryotic aminoacyl tRNA synthetase having a specificity to the natural type amino acid and a tRNA gene capable of binding to the natural type amino acid in the presence of the eukaryotic aminoacyl tRNA synthetase; and (b) knocking-out a gene encoding an endogenous aminoacyl tRNA synthetase having a specificity to the natural type amino acid and an endogenous tRNA gene capable of binding to the natural type amino acid in the presence of the endogenous aminoacyl tRNA synthetase, both of the genes being present inherently in the prokaryotic cell. |
