abstract The present invention relates to novel compositions, methods and tools for modulating nuclear receptor function. More particularly, the invention discloses the identification of the crystal structure of nuclear receptors and the atomic interface with ligands thereof. The invention allows the design, identification or selection of novel candidate modulators of nuclear receptors, which may be used in the pharmaceutical industry. The invention also discloses novel nuclear receptor constructs, recombinant vectors, cells and methods for producing the same in high amounts, as well as their use to produce antibodies, ligands or crystals thereof. The invention is particularly suited for developing modulators of mineralocorticoid receptors, which may be used for treating various disorders, including hypertension.
