abstract A scan microscope, especially a confocal scan microscope, comprises a light source (1) for producing an illumination light (2), a lens system (17, 21) for guiding the illumination light (2) to a sample plane of a sample (19), and a position detector (28) for measuring the lateral deflection, caused by the sample (19), of an illumination light passing through the sample (19). In order to allow for an especially high dissolution, at least one lens system (17, 21) is arranged on each side of the sample plane and at least two lens systems (17, 21) which are opposite each other in relation to the sample plane are directed towards each other in such a manner that the illumination light (2) guided through the lens (17, 21) towards the sample (19) can be collected by the opposite lens system (21, 17) and guided to the position detector (28) as the detection light. The invention also relates to a phase contrast microscopy method using the inventive scan microscope.
