abstract A method for assessing assimilable organic carbon (AOC) levels and/or investigating the AOC-inoculum-relation and/or optimizing the method parameters, and/or for determining the microbial growth inhibition ability of a substance, said method comprising the steps of a) providing a water sample having known or unknown AOC level, b) distributing it into at least one set of at least triplicate vials, each set being intended for generating one data point, c) inoculating each vial, d) incubating the sets of inoculated vials, each set at a chosen incubation temperature, e) sampling and determining the growth of the inoculated cells in each vial after at least one incubation time, said determination of the growth of the microbial cells being performed by (i) fluorescence-staining of the microbial cells, (ii) enumerating the stained cells by means of a flow cytometer, f) generating an AOC-dependent and/or inoculum-dependent information is described. Some advantages of the method are that it can be automated, and that AOC levels can be obtained at stationary phase or at pre-stationary phase.
